Development of Dot – Enzyme Linked Immunosorbent Assay for Detection of Antibodies to Infectious Bursal Disease, Hydropericardium Syndrome and Chicken Anaemia Viruses

چکیده
A rapid and sensitive dot-ELISA test was developed for the simultaneous detection of antibodies against infectious bursal disease (IBD), hydropericardium syndrome (HPS) and chicken anaemia (CAV) viruses in a single serum sample. The developed dot-ELISA was compared with the virus neutralization test (VNT). For IBD and HPS, viruses isolated and purified from field outbreak materials were used while VP1 ORF3 recombinant protein expressed in DH5α E.coli was used for CAV as coating antigens. Sera samples from 302 breeder chicken were screened for antibodies to the three viruses. A total of 195, 205 and 104 serum samples were found positive for IBDV, HPSV and CAV respectively. By comparing with VNT sensitivity, specificity and accuracy of the test were calculated. The dot-ELISA test was found to be highly sensitive, specific with a K value of 0.99 and hence could be very useful for serological diagnosis of IBDV, HPS and CAV in a single serum sample.

Designing an enzyme-linked immunosorbent assay for detection of anti-penicillin antibodies levels in various species of animals in Khuzestan province, Iran
The penicillin allergy is being increasingly recognized as a significant public health problem. Immunological responses to penicillin and other beta-lactam antibiotics are classified as immediate and non-immediate responses. This research aimed to develop an enzyme-linked immunosorbent assay (ELISA) for the detection of the reactive antibody value against penicillin in various species of animals...

An indirect enzyme–linked immunosorbent assay (ELISA) was developed for screening of antibody to avian infectious bronchitis virus (IBV). Antigen was prepared from whole-purified IBV Massachusetts serotype (BR 801 strain). Optimum dilution with minimum background for antigen concentration, rabbit anti-chicken conjugate and sera in developed ELISA was determined 0.1μg/ml, 1:3000 and 1:100, respe...

Nucleocapsid (N) protein of infectious bronchitis virus (IBV), one of the viral structural proteins, inducesstrong antibody response in natural infection. In this study, a simple, recombinant N protein-based dot-blottest was developed to serologically examine chicken serum samples for the presence of IBV antibody.Initially, 72 serum samples were tested for the presence of IBV antibody using a c...

Newcastle disease virus (NDV), avian influenza virus (AIV), and infectious bronchitis virus (IBV) are the most prevalent viral pathogens in the Iranian poultry industry. This study aimed to reveal the presence of these viruses in the backyard chickens in Ahvaz, located in the Southwest of Iran. A total of 100 chickens with respiratory signs and mortality were examined by taking the blood sample...
